How NAD+ and NMN Work

NAD+ (Nicotinamide Adenine Dinucleotide) is a coenzyme found in every living cell. Your cells use it in hundreds of enzymatic reactions, including energy metabolism, DNA repair and cellular signalling.

What the Research Shows

NAD+ and NMN are among the most actively studied compounds in longevity and ageing research:

  • Energy metabolism: NAD+ is essential for mitochondrial function and ATP production — the primary energy currency of cells. (Verdin, Science, 2015;350:1208–1213)
  • Sirtuin activation: NAD+ is a required co-substrate for sirtuins (SIRT1–7), proteins involved in DNA repair and metabolic regulation. (Imai & Guarente, Trends in Cell Biology, 2014;24(8):464–471)
  • NMN in humans: A randomised, placebo-controlled trial found that NMN supplementation raised muscle insulin sensitivity. The full finding is set out below. (Yoshino et al., Science, 2021)

The NMN human trial, in full

Yoshino and colleagues ran the first randomised, placebo-controlled trial of NMN in humans: 250mg daily for ten weeks in 25 postmenopausal women with prediabetes and obesity. Insulin-stimulated glucose disposal, measured by hyperinsulinemic-euglycemic clamp, rose about 25% on NMN and did not change on placebo. Muscle insulin signalling (phosphorylation of AKT and mTOR) increased on NMN as well.
